Structure-guided design methods start from a naturally occurring motif. The motif is then stabilized and improved upon through mutagenesis, crosslinking, or scaffolding. Often, a combination of methods is required to obtain peptides with desired affinities. Note that in some cases, a de novo designed peptide can be built around an already existing motif using some of these approaches (more details in Section 3).
